STATIN

STATIN selectively and competitively inhibits the hepatic enzyme HMG-COA reductase i.e. responsible for converting HMG-COA to mevalonate in the cholesterol biosynthesis pathway, resulting in a subsequent decrease in hepatic cholesterol levels.

Atorvastatin 10/20mg Plasma concentrations achieved in 1 to 2 hours. Volume of distribution: 381L Protein binding: >98% bound to plasma proteins Half life: 14 hours but half-life of HMG-CoA inhibitor activity is 20-30 hours due to longer lived active metabolites

The recommended starting dose is 10 and 20 mg once daily. Patient requiring a large reduction in LDL-C started dose at 40mg once daily.
