Premature Ejaculation

The drug's mechanism of action is thought to be related to inhibition of neuronal reuptake of serotonin and subsequent potentiation of serotonin activity. Experimental evidence indicates that serotonin (5-HT), throughout brain descending pathways, exerts an inhibitory role on ejaculation

Absorption: Rapidly absorbed. peak plasma concentrations are reached within 60–80 min and has an absolute bioavailability of 42%. Distribution: It is 99% protein-bound and has a volume of distribution of 2.1 L/kg. Metabolism: Extensively metabolized by many pathways to give ≈ 40 metabolites. Despite their multiplicity, the dapoxetine metabolites are essentially inactive or are found at pharmacologically insignificant concentrations. Elimination: Rapidly and almost completely eliminated from the body within 24 h, showing minimal accumulation after daily dosing.

Starting dose for all patients (age 16-64 yrs) is 30 mg, taken as needed approximately 1 to 3 hours prior to sexual activity. If the individual response to 30 mg is insufficient the dose may be increased to a maximum recommended dose of 60 mg taken as needed.
