VALID-ER dissociates to the valproate ion in the gastrointestinal tract and its activity in epilepsy is related to increased brain concentrations of gamma-aminobutyric acid (GABA).

Bioavailability: approximately 90%. Time to maximum plasma valproate concentrations (Cmax): 4 to 17 hours. Plasma protein binding: 10% at 40mcg/mL to 18.5% at 130 mcg/mL. Protein binding of valproate is reduced in the elderly, in patients with chronic hepatic diseases, in patients with renal impairment, and in the presence of other drugs (e.g. aspirin). Mean plasma clearance:  0.56 L/hr/1.73m2 Volume of distribution: 11L/1.73m2 Mean terminal half-life: 9 to 16 hours following oral dosing regimens of 250 to 1000 mg.

The dose of VALID-ER will be different for different patients. For Mania Adults: Dose is based on body weight and must be determined by doctor. At first, the usual dose is 25mg per kg of body weight once a day which may be increase. However, the dose is usually not more than 60mg per kg of body weight per day. Children: Use and dose must be determined by doctor. For Migraine Adults: At first, 500mg once a day for 1 week which may increase as needed. However, the dose is usually not more than 1000mg per day. Children: Use and dose must be determined by doctor. For seizures Adults and children 10 years of age or older: Dose is based on body weight. At first, the usual dose is 10 to 15 mg per kg of body weight per day which may increase gradually every week by 5 to 10 mg per kg of body weight if needed. However, the dose is usually not more than 60mg per kg of body weight per day. Children younger than 10 years of age: Use and dose must be determined by doctor.
